TREM2

TREM2，即髓樣細(xì)胞觸發(fā)受體2，它是一種主要在中樞神經(jīng)系統(tǒng)的小膠質(zhì)細(xì)胞中表達(dá)的跨膜受體蛋白。TREM2通過其胞外免疫球蛋白結(jié)構(gòu)域識別多種配體，包括病原體相關(guān)分子模式（PAMPs）和損傷相關(guān)分子模式（DAMPs）。其作用機(jī)制涉及與配體結(jié)合后激活下游信號通路，主要通過與DAP12結(jié)合，激活Syk激酶，進(jìn)而觸發(fā)包括PI3K、PLCγ和RAS/MEK/ERK等在內(nèi)的多種信號傳導(dǎo)途徑。這些信號通路影響小膠質(zhì)細(xì)胞的吞噬功能、細(xì)胞存活、代謝平衡和炎癥反應(yīng)。在生物學(xué)意義上，TREM2對維持中樞神經(jīng)系統(tǒng)穩(wěn)態(tài)、調(diào)節(jié)免疫反應(yīng)和參與神經(jīng)退行性疾病的發(fā)病機(jī)制具有重要作用。TREM2信號的異常激活與阿爾茨海默病等神經(jīng)退行性疾病的發(fā)生發(fā)展密切相關(guān)，是當(dāng)前神經(jīng)科學(xué)研究的熱點(diǎn)之一。

Immunofluorescence staining of HepG2 cells with CSB-PA024405LA01HU at 1:166, counter-stained with DAPI. The cells were fixed in 4% formaldehyde, permeabilized using 0.2% Triton X-100 and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4°C. The secondary antibody was Alexa Fluor 488-congugated AffiniPure Goat Anti-Rabbit IgG(H+L).
