SIRT7
SIRT7，全稱為NAD+依賴性組蛋白去乙酰化酶SIRT7，是sirtuin家族的一員，該家族在調(diào)節(jié)多種生物學(xué)過(guò)程中發(fā)揮作用，包括基因組穩(wěn)定性、代謝途徑、應(yīng)激反應(yīng)和腫瘤發(fā)生。SIRT7蛋白主要位于細(xì)胞核，是一種高度特異性的H3K18Ac（組蛋白H3的第18位賴氨酸殘基乙酰化）去乙酰化酶。近年來(lái)的研究發(fā)現(xiàn)，SIRT7蛋白可以通過(guò)多種途徑參與調(diào)控核糖體RNA轉(zhuǎn)錄、細(xì)胞代謝、細(xì)胞應(yīng)激以及DNA損傷修復(fù)等生理過(guò)程。
SIRT7還與衰老、心臟疾病及脂肪肝等密切相關(guān)。特別是SIRT7在多種腫瘤如肝癌、胃癌、乳腺癌、膀胱癌、結(jié)直腸癌、胰腺癌和頭頸鱗狀細(xì)胞癌等發(fā)生發(fā)展中起著重要的調(diào)節(jié)作用。越來(lái)越多的證據(jù)表明，SIRT7蛋白的基本細(xì)胞程序功能對(duì)于致癌演變以及腫瘤生物學(xué)有重要的影響，因此SIRT7蛋白有望成為表觀遺傳藥物治療新靶點(diǎn)。
