MICA
MICA，全稱為人類MHC-I類分子鏈相關(guān)基因A蛋白，是一種主要在腫瘤細(xì)胞表面表達(dá)的免疫原性蛋白。該蛋白常見別名包括MHC class I chain-related antigen A、CD155等。MICA蛋白與NKG2D受體結(jié)合，激活NK細(xì)胞和CD8+ T細(xì)胞，從而殺傷腫瘤細(xì)胞。在腫瘤免疫逃逸中，MICA蛋白的表達(dá)下調(diào)或脫落是常見機(jī)制。針對MICA蛋白的藥物研發(fā)已成為腫瘤免疫治療的重要方向。目前，已有多種靶向MICA蛋白的單克隆抗體進(jìn)入臨床試驗(yàn)階段，如CLN-619和Genentech的1D5V11抗體。這些藥物通過阻斷MICA蛋白與NKG2D受體的結(jié)合，或抑制MICA蛋白的脫落，從而增強(qiáng)NK細(xì)胞和T細(xì)胞對腫瘤細(xì)胞的殺傷作用。此外，還有研究探索MICA蛋白在其他疾病中的作用，如自身免疫性疾病和病毒感染等。MICA蛋白的研究為腫瘤免疫治療提供了新的思路和策略，有望為癌癥患者帶來新的治療選擇。

IHC image of CSB-RA574653A0HU diluted at 1:80 and staining in paraffin-embedded human small intestine tissue performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a Goat anti-rabbit polymer IgG labeled by HRP and visualized using 0.49% DAB.
